PRISM In Vivo Imaging Systems Now Ship Fully Integrated with Kent Scientific’s SomnoFlo

MediLumine is pleased to announce that Kent Scientific, makers of the SomnoFlo low-flow anesthesia system, is now an authorized distributor of PRISM in vivo imaging systems.

PRISM systems now ship fully integrated with SomnoFlo, giving laboratories a complete, turnkey small animal imaging workflow out of the box. The PRISM platform has been cited in more than 40 peer reviewed publications.

Low-Flow Anesthesia with No Compressed Gas

SomnoFlo is a low-flow electronic vaporizer that runs on ambient air and draws isoflurane directly from the bottle. As a result, no compressed gas cylinder is required in the imaging suite.

The vaporizer offers 0.1 percent precision with flow rates from 50 mL/min. Because the benchtop PRISM and its anesthesia suite operate as one system, the setup can move between laboratories as a single unit. The configuration is also safer for both animals and technicians.

Bioluminescence and Fluorescence Imaging of up to 10 Mice

The PRISM BIO-BOX is designed for bioluminescence imaging (BLI) and fluorescence imaging (FLI) of up to 10 mice within a 25 cm field of view.

The system uses a 9 MP sensor (3000 x 3000 array) with a 3.76 µm native pixel. It can zoom to a 4 cm field of view for 13.3 µm/pixel object space sampling. Dark current is below 0.001 e-/pixel/s, and a full 9 MP frame is read in under a second with live preview. An extended dynamic range mode provides 2.5x additional dynamic range.

Sensitivity is verified against a NIST traceable source at the BLI autoluminescence threshold. A heating pad maintains animal temperature between 20 and 45°C.

Acquisition and image analysis are handled by the PYRAMID software suite, which includes unlimited licenses. The benchtop unit measures 61 x 61 x 91 cm and runs on a standard 120 V / 15 A outlet.

Uniform Illumination and Flexible Filter Options

For fluorescence imaging, a dual broadband white light source with heads at the top left and top right of the chamber, each fitted with collimating optics, provides uniform illumination across the full 25 x 25 cm field.

The base configuration offers 24 excitation and emission combinations (6 x 4), expandable to 360 (18 x 20). These are selected from an inventory of 14 excitation and 7 emission filters spanning 460 to 830 nm. Acquisition sequences can be automated for multicolor FLI and BLI.

Case Study: mRNA LNP Biodistribution with BLI

Bioluminescence images of five mice at 6, 24 and 72 hours after LUC+ mRNA LNP injection acquired on the PRISM BIO-BOX

An example case study shows the PRISM BIO-BOX applied to mRNA lipid nanoparticle (LNP) research. Five mice were imaged dorsally at 6, 24, and 72 h following injection of LUC+ mRNA LNP supplied by RIBOPRO (Netherlands).

Mice 1 and 2 received intravenous injections, mice 4 and 5 received subcutaneous injections, and mouse 3 served as a control. Regions of interest 1 through 5 were placed over the liver and spleen, with ROI 6 used as background.

Images were acquired at exposures of 1, 2, and 2.5 min and displayed on a common radiance scale (ph/s/cm²/sr), allowing signal to be compared across animals, administration routes, and time points.

Optional SWIR / NIR-II Imaging

An optional SWIR / NIR-II module adds a second InGaAs camera covering 900 to 1700 nm. It is integrated within the same enclosure via a dichroic beamsplitter with 1000 and 1300 nm longpass filters.

In one example, intravenous injection of a pegylated AIE nanoparticle excited with the PRISM SWIR 785 nm laser revealed detailed vascular structures. Signal was also evident in superficial skin lesions and in clearance through the bladder, highlighting both biodistribution and excretion pathways.

3D Imaging and Motorized Stage Options

3D trans-fluorescence is available using laser transillumination with depth reconstruction. It is configured with one laser at 635, 660, 730, or 785 nm and is expandable to four. A 3D profile scanner can also be used for 3D BLI.

A motorized vertical stage provides automated height control with autofocus for a variable 4 to 25 cm field of view.

BSL3 Isolation and Contrast Agents

For facilities with elevated biosafety requirements, an optional 10 mouse isolation chamber is available for BSL3 environments, and HEPA filters can be easily added to the unit.

Contrast agents for BLI, FLI, and SWIR imaging are also available, including Luciferin Potassium Salt, Fenestra ICG, and Vascular Tracker1010.

The VIVA™ physiological monitoring platform is a compact monitoring system designed to integrate seamlessly with the PRISM in vivo imaging platform or operate as a standalone unit. It provides precise real-time tracking of ECG and body temperature in small animals. Built-in paw electrodes capture clean, stable cardiac signals, while a precision rectal probe continuously measures core body temperature. The system’s magnetic docking interface allows effortless transition between benchtop monitoring and imaging inside the PRISM BIO-BOX™.
