GadoSpin™ F

Description

Product Overview

GadoSpin™ F is a specialized MRI contrast agent developed for in vivo imaging of atherosclerotic plaques. Its amphiphilic design includes a hydrophobic targeting moiety that facilitates high protein binding and selective accumulation within atherosclerotic lesions. This results in robust T1-weighted enhancement of plaque burden and vascular structures in preclinical models.

Schematic diagram of GadoSpin F:

Top: Accumulation of GadoSpin F in atherosclerotic plaque of a WHHL rabbit 6 hours (left) and 24 h (right) post injection, as measured by as measured by T1-weighted MRI (TFL IR, 1.5 T).

Bottom: Axial Flash MR images of an ApoE-/-mouse pre (left) and 24 hours post (right) injection of GadoSpin F. The signal intensity of the descending aorta (red arrows) increases due to GadoSpin F accumulation in atherosclerotic plaque.

Research Applications

GadoSpin™ F enables quantitative and qualitative assessment of atherosclerotic plaques in small animal models. The agent’s long blood pool residence allows for additional angiographic imaging during the same session. Beyond plaque imaging, GadoSpin™ F can be utilized for selective visualization of lymph nodes and sites of inflammation due to its targeted enrichment properties.

Imaging Characteristics

The agent demonstrates high relaxivity at physiological temperature and field strength, with r1 of 18 L mmol-1 s-1 and r2 of 31 L mmol-1 s-1 in plasma at 37 °C, and r1 of 15 L mmol-1 s-1 and r2 of 20 L mmol-1 s-1 in water. Its molecular weight is approximately 1,300 g mol-1. These characteristics enable clear T1-weighted positive contrast in both murine and rabbit models of atherosclerosis.

Advantages for Preclinical MRI Studies

GadoSpin™ F allows accurate visualization and quantification of plaque burden with minimal procedural complexity. Its selective targeting and extended blood pool phase support longitudinal studies and facilitate complementary angiographic imaging. The agent’s amphiphilic nature and high protein binding contribute to stable signal enhancement in vascular and inflammatory tissues.

Preclinical Imaging Validation

In preclinical studies, GadoSpin™ F accumulates selectively in atherosclerotic plaques. In WHHL rabbits, significant signal enhancement is observed at six and twenty-four hours post injection. In ApoE-/- mice, T1-weighted MRI demonstrates increased signal intensity in the descending aorta, highlighting plaque regions.
